She was born in August 1987 to a Punjabi Sikh family in Delhi. Her father Dilmohan Singh Pannu ran a paper mill; her mother Nirmal a homemaker. She finished a B.Tech. in computer science from Guru Tegh Bahadur Institute of Technology, affiliated to GGSIPU, and worked briefly as a software engineer before deciding the cubicle and the cafeteria were not, in fact, going to be her life.

Modelling in Delhi led to a Telugu film, K. Raghavendra Rao's Jhummandi Naadam (2010) opposite Manchu Manoj. Tamil and Telugu cinema absorbed her for the next four years — Aadukalam (2011), Mr. Perfect (2011), Vandhaan Vendraan (2011), Mogudu (2011) — before David Dhawan's Chashme Baddoor (2013), a Hindi remake, gave her her first Hindi feature, and Neeraj Pandey's Baby (2015) — as a Mossad-trained operative opposite Akshay Kumar — was her first Hindi crossover.

Pink (2016), a courtroom drama about three Delhi women accused of soliciting after a night out, was the role that fixed her trajectory. The film grossed over Rs 1 billion against a Rs 220-million budget; its closing monologue — the one about consent — became a recurring exhibit in Indian sexual-harassment workshops for the next half decade. Naam Shabana (2017) and Mulk (2018) consolidated the type: the urban woman in a system designed to disbelieve her.

Anubhav Sinha's Thappad (2020), in which her character files for divorce after her husband slaps her at a party in front of his colleagues, was the most precise of her message-driven films. The film opened on 28 February 2020 and was effectively shut down by the COVID lockdown within twelve days; it endured anyway, finding a second life on Amazon Prime Video and becoming the year's most-discussed Hindi script.

Saand Ki Aankh (2019), in which she and Bhumi Pednekar played sexagenarian sharpshooters Chandro and Prakashi Tomar from the village of Johri in Uttar Pradesh, drew accusations of casting older characters with younger actresses; Pannu defended the choice publicly and at cost, and used the platform to talk about the actual Tomars' shooting range.

Mission Mangal (2019) and Rashmi Rocket (2021) — as a Kutchi athlete subjected to a sex-determination test by an Indian sports federation — extended the range. Looop Lapeta (2022), a Hindi remake of Tom Tykwer's Run Lola Run, was an experiment that did not entirely succeed; her producer-side bet on its risks did succeed.

She co-founded the production company Outsiders Films in 2021 with Pranjal Khandhdiya, formerly of Reliance Entertainment, with the explicit mandate of producing female-led genre cinema. The company's first film, Blurr (2022) — a Hindi remake of the Spanish thriller Julia's Eyes — was a calling-card; subsequent slate includes the still-in-development Woh Ladki Hai Kahaan.

She married the Danish badminton player Mathias Boe in March 2024 at the Suján Rajmahal Palace in Udaipur, a marriage she had refused, characteristically, to confirm until the day after the ceremony. The couple had been together for almost a decade.

Phir Aayi Hasseen Dillruba (2024) for Netflix, Dunki (2023) opposite Shah Rukh Khan in Rajkumar Hirani's emigrant drama, and the recently shot Khel Khel Mein (2024) make up the late-career commercial portfolio. The message-driven films, however, remain the spine; nothing in her output suggests she intends to abandon them.
