Following his doctoral defense, Poénaru moved to the United States to conduct research, spending four years divided between appointments at Harvard University and Princeton University. He eventually returned to France in 1967. Throughout his tenure at the University of Paris-Sud, he focused his teaching and research on low-dimensional topology, ultimately supervising students such as Jean Lannes.

Poénaru maintained a multi-decade interest in the Poincaré conjecture. His engagement with the problem began as early as 1957. By December 19, 2006, he submitted a preprint to the arXiv titled On the 3-Dimensional Poincaré Conjecture and the 4-Dimensional Smooth Schoenflies Problem, asserting a completed proof.
