In 1990, Mielke achieved habilitation at the University of Stuttgart, focusing his research on Hamiltonian and Lagrangian flows on center manifolds, specifically applied to elliptic variational problems. This foundational work preceded his 1992 appointment as a professor at the University of Hannover. By 1999, he returned to the University of Stuttgart to join the Institute for Analysis, Dynamics, and Modeling.

Since 2004, Mielke has served as a full professor of applied analysis at the Humboldt-Universität zu Berlin. Simultaneously, he holds the position of head of the research group on partial differential equations at the Weierstrass Institute for Applied Analysis and Stochastics in Berlin.
