Early Development and Ajax
Vermaelen began his youth career in his home country with Germinal Ekeren, which later merged to become Germinal Beerschot. He subsequently joined the youth academy of the Dutch club Ajax in 2000. He secured his professional debut on 15 February 2004 during a 2-0 victory against FC Volendam. Following a loan spell at RKC Waalwijk, he returned to Ajax, eventually serving as club captain for the remainder of the 2008-09 season.

In June 2009, Vermaelen joined Arsenal on a four-year contract. He debuted in the Premier League on 15 August 2009, scoring during a 6-1 win over Everton. During his first season, he was included in the PFA Premier League Team of the Year. He assumed the role of club captain in August 2012, though his first-team participation later decreased due to the defensive partnership formed by Per Mertesacker and Laurent Koscielny.
International and Late Career
Vermaelen made his international debut for Belgium in 2006. His international participation included the 2008 Olympics. Following his time in Spain with Barcelona, he transitioned to the Japanese club Vissel Kobe in 2019. He officially retired from professional football in 2021.
